摘要
目的探讨胞外-5′-核苷酸酶(5′-NT,CD73)在大鼠肝脏缺血预处理中的作用。方法建立大鼠肝脏热缺血再灌注模型。缺血预处理采用肝脏缺血3min,再灌注3min,反复重复此过程4次。经30min缺血后再灌注,利用Westernblot检测不同时间点CD73在大鼠肝脏组织中的表达,HE染色显示肝脏病理组织学变化,酶学方法检测血清酶学指标(LDH、AST、ALT)变化。结果CD73的蛋白表达在大鼠肝脏缺血预处理后明显升高。药物抑制CD73的功能可以消除缺血预处理的作用,可溶性5′-核苷酸酶可以保护肝脏缺血损伤。结论CD73在大鼠肝脏缺血预处理的保护机制中发挥重要作用。可溶性5′-核苷酸酶可能用于肝脏缺血损伤的治疗。
Objective To investigate the function of ecto-5'-nucleotidase (5'-NT, CD73 ) in ischemie preconditioning (IP) of rat livers. Methods After the models of sham-operation and hepatic ischemie were established in rats,IP was performed with 4 cycles of 3 minutes' ischemia followed by 3 minutes' reperfusion. After 30 minutes of ischemic repeffusion,the expression of CD73 protein in rat livers at different time point was detected by Western blot, hepatic histopathologieal changes was observed with HE staining, and the changes of LDH, AST and ALT were analysed by serum enzymatic measurement. Results The expression of CD73 protein significantly increased after ischemic preconditioning in rat livers. Pharmacological inhibition of CD73 could abolish the function of ischemie preconditioning. Soluble 5'-nucleotidase could protect the liver damnifieation of ischemic reperfusion. Conclusion CD73 might play an important role in the protective mechanism of ischemie preconditioning for rat liver. Soluble 5'-nucleotidase could be a potential therapy for hepatic isehemia.
