摘要
目的:建立侵袭性感染丝状真菌的分子鉴定方法。方法:收集上海市公共卫生临床中心于2007年12月至2009年3月间分离的侵袭性感染丝状真菌26株,用真菌通用引物Its1和Its4采用聚合酶链反应(PCR)扩增核糖体RNA基因序列,对PCR产物进行DNA序列测定,确定丝状真菌的菌种。结果:26株侵袭性感染丝状真菌中鉴定出黄曲霉5株,马内菲青霉4株,土曲霉、草酸青霉各3株,烟曲霉、禾谷镰刀菌各2株,日本曲霉、沙门柏干酪青霉、轮枝菌、塔宾曲霉、小麦壳针孢菌、球毛壳菌、李氏木霉各1株。结论:使用真菌核糖体RNA基因序列分析可快速明确传统方法难以鉴定的丝状真菌菌种。
Objective To identify filamentous fungi involved in invasive mycoses by using PCR and DNA sequencing. Methods Twenty-six clinical isolates of filamentous fungi were collected during December 2007 to March 2009 from Shanghai Public Health Clinical Center. The rRNA gene sequence of filamentous fungi was amplified by PCR using fungal universal primers Itsl and Its4. The PCR products were sequenced to identify the fungal species. Results In the 26 clinical isolates, 5 strains ofAspergillus flavus, 4 strains ofPenicillium marneffei, 3 strains each ofAspergilluz terreus and PeniciUium oxalicum, 2 strains each of Aspergillus fumigatus and Fusarium chlamydosporum, and 1 strain each of Aspergillus japonicas, PeniciUium camemberti, Verticillium sp., AspergiUus itaconicus, MycosphaereUa laterolis, Chaetomium globosurn and Hypocrea lixii were identified. Conclusions The rRNA gene sequencing of filamentous fungi could be used for rapid strain identification to identify filamentous fungi difficulty to be identified by conventional methods.
