一种研究脑白质空泡样改变SD大鼠模型的建立 被引量:1

Establishing a Model of SD Rat for Investigating Sponge-like Changes in the White Matter of the Brain

摘要 目的用双环己酮草酰二腙(cuprizone)建立SD大鼠脑白质空泡样改变动物模型并观察病变部位特征。方法将cuprizone按10mg/100g体重/日经口腔灌注,分别在4周和6周用H.E染色观察脑白质病理改变,采用Hoechst33258染色法检测细胞凋亡。结果实验大鼠脑白质于4周时无明显空泡样改变形成,但在脑干及大脑深层白质可见阳性凋亡细胞;6周时分别在脑干(7/10)和大脑白质深层(4/10)可见空泡样改变形成,仍可检测出凋亡细胞。结论双环己酮草酰二腙口腔灌注法可在SD大鼠建立脑白质空泡样改变模型,细胞凋亡早于空泡形成。提示细胞凋亡可能是引起白质空泡样改变的原因之一,并可以成为一种研究细胞凋亡与空泡样改变相互关系的实验动物模型。 Objective To explore the method of using cuprizone for building the model of sponge change in white matter of SD rats and determine the feature of lesions. Methods SD rats were poured with euprizone 10mg per 100g weight per day. Pathological changes of white marten in the brain were demonstrated wiht H. E stained brain sections and apoptosis of the brain sections was detected by Hoechst33258 staining method respectively. Results Spongiform was not discovered in white matter of the brain at 4 weeks but some apoptosis was found in brain stem and eddp white matter of the brain. Vacuolizations in corpus callosum and deep white matter of cerebrum were observed in 6 week groups while apoptosis still existed. Conclusion The method of pouring with cuprizone in SD rats be used as a model for spongiform of white matter. Apoptosis was earlier than the spongiform. The apoptosis was supposed to be one of the cause inducing the spongiform of the white matter in the brain.
