摘要
目的探讨骨髓内皮祖细胞移植对单侧输尿管梗阻大鼠肾小管周毛细血管重建的影响。方法应用密度梯度离心法分离骨髓单个核细胞,内皮祖细胞选择性培养液培养后移植到单侧输尿管梗阻大鼠。应用免疫组织化学半定量单侧输尿管梗阻大鼠肾间质微血管密度的改变。应用Westernblot检测血管生成素-1(Ang-1)和血管内皮生长因子(VEGF)表达。结果单侧输尿管结扎模型(UUO)组在术后14、21d肾小管周毛细血管指数较假手术组明显下降(P<0.05),毛细血管变形表现为明显的扭曲、萎缩或扩张。移植组在14、21d肾小管周毛细血管指数(PCI)显著低于假手术组(P<0.05),但高于同期较UUO组(P<0.05)。与假手术组比较,UUO组、移植组Ang-1、VEGF表达第7天显著增加,随后迅速下降,而移植组各时相点均高于同期UUO组(P<0.05)。结论内皮祖细胞移植以旁分泌、自分泌机制参与单侧输尿管梗阻大鼠肾小管周毛细血管重建。
Objective To research the effect of endothelial progenitor cells(EPCs) transplantation on reconstitution of peritubu lar capillaries in UUO rats. Methods Mononuclear cells(MNCs) in rat bone marrow were isolated by density gradient centrifugation and were cultured according to previously described techniques, then transplanted by direct renal artery injection. Peritubular capillaries density was evaluated by expression of immunohistochemistry JG12. The protein expression of vascular endothelial growth gactor(VEGF) and angiogenin(Ang-1) was assessed by Western blot method. Results In transplantation group peritubular capillary index on 14d and 21d in transplantation was significantly lower than that in Sham group, and higher than that in UUO group(P〈0.05). Compared with sham group,expression of Ang-1 and VEGF increased on 7d(P〈0.05) ,but quickly decreased, and in transplantation group the increase was higher and longer(P〈0.05). Conclusion EPCs participated in peritubular capillaries reconstitution by autocrine and paracrine.
