摘要
目的探讨肿瘤坏死因子-α(TNF-α308位点)基因多态性与宁夏汉族煤工尘肺(CWP)患者的关系。方法对宁夏神华煤业集团所属煤矿已确诊的110例煤工尘肺病例与110例接尘对照者和110例非接尘对照者进行研究(本文研究对象均为汉族),采用聚合酶链反应-限制片断长度多态(PCR-RFLP)分析TNF-α基因中-308G/A多态性位点。结果煤工尘肺组[(48.30±13.16)岁]与煤尘接触组[(43.86±4.20)岁]和非煤尘接触组[(41.73±5.45)岁]间年龄差异有统计学意义(P<0.01);煤工尘肺组和煤尘接触组之间接尘工龄(t=0.47,P=0.64)、主掘进工和主采煤工所占比例、吸烟烟龄等差异均无统计学意义;TNF-α-308G/A基因型在煤工尘肺组和煤尘接触组及非煤尘接触组中所占比例分别为35.45%、23.60%、18.20%;等位基因频率A在煤工尘肺组和煤尘接触组及非煤尘接触组中所占比例分别为20.46%、13.6%、10.01%;TNF-α-308G/A基因型与等位基因频率分布差异有统计学意义。结论肿瘤坏死因子-α基因型-308位点可能与宁夏煤工尘肺病的发生有关。
Objective To investigate the relationship between gene polymorphisms of Tumor necrosis factor-αand coal worker pneumoconiosis.Methods In a prospective study of 110 patients with CWP and 110 controls,TNF-α-308G/A genotypes were measured using PCR-restriction fragment length polymorphism.Results The average age of coal worker pneumoconiosis group was 48.30±13.16,and they were 43.86±4.20 and 41.73±5.45 yrs in coal dust exposure group and nondust exposure group respectively.No significantly difference was found Dust length of service, the main tunneling work, age of smoking cigarettes. TNF-αin CWP patients were significantly higher than that in controls ( P 〈 0.05). TNF-α-308G / A genotype accounted by 35.45% ,23.60%, 18.20% in coal worker pneumoconiosis group, coal dust exposure group and non-dust exposure group respectively. Allele frequency of A was 20.46%, 13.6%, 10.01% in coal worker's pneumoconiosis group ,dust exposure group and non-coal dust exposure group respectively. Significant difference between CWP patients and controls was found in frequencies of TNF-α-308G/A genotypes and alleles (P 〈 0.05). Conclusion TNF-α may be related to CWP patients in Ningxia.
