摘要
目的研究蝙蝠葛酚性碱(PAMD)对BxPC-3裸鼠的抑瘤作用及对肿瘤新生血管bFGF表达的影响,进一步探讨其作用机制。方法建立BxPC-3裸鼠的动物模型,检测蝙蝠葛酚性碱对转移瘤的抑制率,同时应用SP免疫组织化学方法检测各组BxPC-3裸鼠肿瘤组织bFGF的表达水平。结果蝙蝠葛酚性碱对BxPC-3裸鼠瘤组织具有抑制作用,高、中、低剂量组抑瘤率分别为34.91%、52.83%、41.51%,与模型对照组比较具有统计学意义(P<0.01);各剂量均能明显降低BxPC-3裸鼠瘤组织bFGF的表达水平。结论蝙蝠葛酚性碱对BxPC-3裸鼠肿瘤具有明显的抑制作用,其机制可能是通过改善裸鼠免疫功能,抑制肿瘤新生血管形成来实现的。
Objective To explore the effects of phenolic alkaloids from menispermum dauricum (PAMD) on tumor inhibitory and expression of basic fibroblast growth factor(bFGF) in BxPC-3 nude mice and to further investigate its protective mechanism for pancreatic carcinoma. Methods The metastatic tumor model of nude mice was estab lished with BxPC-3 cell line. After PAMD treatment, the inhibition rate of tumor proliferation was detected and immunohistochemistry was carried out to confirm the expression of bFGF in nude mice. Results PAMD could inhibit the tumor growth in nude mice of BxPC-3 cell line and the inhibition rate with high, middle and low dose PAMD was 34. 91%,52.83%, 41.51%, respectively( vs control group, P〈 0. 01 ). Meanwhile, PAMD could apparently inhibit the expression of bFGF in the tumor. Conclusion PAMD shows conspicuous inhibitory action on the tumor growth of nude mice,which possibly due to its potency to improve immune function and inhibit tumor angiogenesis in nude mice.
