摘要
目的研究受体胸腺内注射Fas抗体对心脏移植大鼠Fas/FasL表达的影响。方法应用大鼠同种异位腹腔心脏移植模型,将24只模型动物随机均分为三组,A组移植前后不给处置,B组给环孢素A,按每日8mg/kg的剂量于术前一天开始口服,C组胸腺注射Fas抗体。观察术后移植心脏局部的Fas及FasL的表达情况(免疫组化法和RT-PCR法)。结果与A组相比,C组移植心脏局部的Fas/FasL基因表达(RT-PCR法半定量结果P<0.05)和蛋白表达减少(免疫组化法P<0.05)。结论受体胸腺内注射Fas抗体是一种有效的抑制心脏移植急性排斥反应的方法,引起移植心肌局部Fas/FasL表达下调。
Objective To investigate the influence of intrathymic injection of anti-Fas monoclonal antibody on Fas/FasL in rats after heart transplantation. Methods Allograft heterotopic heart transplantation models of rats were utilized. Twenty four rats were randomly and evenly divided into three groups. Group A had no special perioperation inter vention. Group B was administrated with cyclosporine a 8 mg/(kg × d) from the day before operation. Group C was injected with Fas antibody in thymus. The expression of Fas/FasL on the local tissue of transplanted heart after operations was observed. Results Compared to group A, group C had less gene expression (RT-PCR, semiquantitative, P〈0. 05) and less protein expression (immunohistochemistry, P〈0. 05) of Fas/FasL on the local transplanted heart. Conclusion Intrathymic injection of anti Fas monoclonal antibody has a immunosuppressive effect on acute rejection reaction and down-regulation on the expression of Fas/FasL in rats after heart transplantation.
