摘要
目的探讨乳腺肿瘤组织中金属蛋白酶组织抑制物TIMP┐3及TIMP┐4mRNA表达与乳腺癌转移的关系。方法以原位杂交方法检测二甲基苯蒽(DMBA)诱发的大鼠乳腺原发瘤组织中此两种mRNA的表达水平,并观察转移组、非转移组、良性肿瘤组的表达意义。结果TIMP┐3及TIMP┐4mRNA均表达于肿瘤间质,乳腺癌非转移组表达水平显著高于转移组及良性肿瘤组。结论TIMP┐3及TIMP┐4mRNA由间质细胞产生,过度表达反映了宿主细胞对肿瘤侵袭的一种应答反应,可能是转移行为的负性调节因子。
Objective:To study the relationship between expression of tissue inhibitor of metalloproteinasesTIMP-3,TIMP-4 mRNAs and metastasis in breast carcinoma. Methods:The expression of TIMP-3 and TIMP-4 mRNAs was examined by in situ hybridization in the 7, 12dimethylbenz(α)anthracene induced breast tumors. Their significance was analysed in the metastatic(M+),nonmetastatic(M-), and benign tumor (BT) groups.Results:It was found that TIMPs signal was localized in stroma,and that the levels of their mRNA expression in M- group were significantly higher than those in M+ group as well as BT group.Conclusion:These findings indicate that TIMP-3 and TIMP-4 mRNAs were produced by stromal cells,and the overexpression of them in breast tumor represented the host response to tumor progression,and might be negative regulators in metastasis.
