男性高血压前期与正常血压及原发性高血压患者血浆比较蛋白质组学研究 被引量:2

Plasma proteomic analysis in male normotensive, prehypertensive and hypertensive subjects

摘要 目的探讨正常血压、高血压前期及高血压患者血浆蛋白质组学差异。方法筛选正常血压者,高血压前期及高血压男性患者各26例,分离血浆,去除高丰度蛋白质,制备蛋白质样品,通过使用二维电泳结合基质辅助激光解析电离飞行时间质谱技术进行各组血浆蛋白质组的分析和鉴定,并通过Western blot进一步进行验证。结果在三组不同血压的患者血浆中存在着22个表达差异的蛋白质点,这些点与18种蛋白质在相关。主要涉及炎症与免疫、脂质代谢、转运蛋白质、凝血和纤溶、细胞增殖和凋亡以及抗氧化蛋白质。结论高血压前期患者血浆蛋白质组与正常血压及高血压患者之间存在明显的差别,高血压组也与正常血压组不同。高血压前期可能是高血压早期的一种表现。 Objective To compare the plasma proteome among male normotensive, prehypertensive, and hypertensive subjects. Methods Plasma proteome was analyzed by two-dimensional electrophoresis combined with MALDI-TOF mass spectrometry in this case-control study among well matched male normotensive, prehypertensive and hypertensive subjects (n = 26 each). Results The results showed that there were 22 differentially expressed protein spots among the protein samples derived from the 3 groups which corresponded to 18 proteins associated with inflammation and immunity, lipid metabolism, transport, coagulation and fibrinolysis, cell proliferation and apoptosis, and antioxidation. Conclusion Proteins were differentially expressed in male subjects with various blood pressure levels.
