摘要
目的探讨放射性损伤骨髓衰竭的机理及防护对策。方法用Northernblot技术检测了60Coγ射线8.0Gy照射小鼠(对照组)及照射后立即给复方活血汤治疗小鼠(中药组)的骨髓有核细胞bcl-2、bax、bcl-xL基因表达。结果放射损伤后第3,7天,对照组骨髓有核细胞bcl-2/bax值减小、bcl-xL不表达。中药组与之相反,bcl-2/bax值明显增大、bcl-xL明显表达。受照后第7天两组脾脏单个核细胞bcl-2/bax值与骨髓有核细胞的表达一致。结论复方活血汤对放射损伤骨髓造血细胞增殖周期调控基因bcl-2、bax、bcl-xL有一定调节作用。
Objective\ To explore the mechanism of and protective strategy againt bone marrow failure caused by radiation injury.Method\ Northern blot technique was used to detect the bcl 2,bax and bcl x L gene expression in murine bone marrow nucleated cells (BMNC) both in Chinese drug treated group which was given per os compound blood activating decoction immediately after irradiation with 8.0 Gy60Co γ rays,and in control group.Results\ On the 3rd and 7th day after irradiation,bcl 2/bax value of BMNC decreased obviously.No bcl x L expression was observed in control group.In Chinese drug treated group,the bcl 2/bax value increased remarkably,while bcl x L expression occurred obviously.On the 7th day after irradiation,the bcl 2/bax value in splenic mononuclear cells was consitstent with that in BMNC in both Chinese drug treated and control groups.Conclusion\ Compound blood activating decoction could have a regulative effect on the expression of cell cycle regulation gene bcl 2,bax and bcl xL in irradiated bone marrow hematopoietic cells and protect the bone marrow hematopoiesis.
