摘要
目的观察乳化异氟烷预处理对在体兔心肌缺血再灌注损伤的影响。方法32只雄性、新西兰白兔随机分为4组,每组8只。缺血对照组(CON组);缺血预处理组(IP组);乳化异氟烷预处理组(EI组);脂肪乳组(INT组)。新西兰白兔均经历缺血30min、复灌180 min过程。记录心率(HR)、平均动脉压(MAP),计算心率收缩压乘积(RPP)。于复灌180 min时抽取颈内动脉血3ml,测定血清肌酸激酶(CK)、乳酸脱氢酶(LDH)活性。复灌180 min后2,3,5-三苯基四氮唑(TTC)法测定心肌梗死面积。结果四组HR、MAP、RPP值组间比较差异无统计学意义(P>0.05),与平衡期末比较,缺血及复灌后四组HR、MAP、RPP值均呈进行性下降(P<0.05)。与CON组和INT组比较,IP组和EI组心肌梗死面积明显降低(P<0.05)。复灌180 min时IP组和EI组血清CK、LDH活性明显低于CON组和INT组(P<0.05)。结论乳化异氟烷预处理对在体兔心肌缺血再灌注损伤具有保护作用。
Aim To investigate the effects in emulsified isoflurane preconditioning against ischemia reperfusion injury in rabbits. Methods Thirty-two male New Zealand white rabbits were randomly divided into 4 groups ( eight for each group) : Control group (CON) ; ischemia preconditioning group(IP) ;Emulsified isoflurane preconditioning group(EI) ;Intralipid group(INT). The rabbits were subjected to 30 rain of myocardial ischemia induced by temporary ligation of the left anterior descending coronary artery(LAD) followed by 180 min of reperfusion. Hemodynamic results were recorded, blood were taken from carotid artery at 180 rain after reperfusion for measuring serum creatine kinase(CK) and lactate dehydrogenase (LDH) levels. Infarct size (IS) was determined by 2,3,5-triphenyltetrazolium (TYC) staining at 180 rain after reperfusion. Results There were no differences in HR, MAP, RPP among four groups ( P 〉 0.05 ), Compared to baseline, HR, MAP, RPP was dropped step by step after ischemia and reperfusion (P 〈 0.05 ). Compared to group CON and INT, myocardial infarct size were lower in group IP and El ( P 〈 0.05 ). At reperfusion 180 min, serum CK and LDH levels in group IP and El were lower compared with group CON and INT(P 〈0.05). Conclusion Emulsified isoflurane preconditioning can provide myocardial protection against ischemia reperfusion injury in rabbits.
