摘要
从碱性土样中经分离筛选得到1株固态发酵产碱性内切聚半乳糖醛酸酶活力较高的菌株。经提酶条件优化得到较优的酶液提取提条件为30倍Na2CO3/NaHCO3缓冲液提取1 h。从形态特征、培养特征、生理生化特征以及分子生物学鉴定结果来看,该菌株属B.gibsonii。
A bacterial strain highly producing alkaline endopolygalacturonase by solid-state fermentation was obtained from a sample of alkaline soil. The recovery ratio of the enzyme from the isolate was the highest after 1 h of extraction in 30 volume of Na2CO3/ NaHCO3 buffer at 30℃. The isolate was confirmed to be a strain of B. gibsonii after identified by colony characteristics, morphological characteristics,physiologica and biochemical characteristics and molecular biology methods.
