过氧化物酶体增殖物激活受体γ在正常人脑和脑星形细胞瘤组织中的表达

Expression of PPARγ in human normal cerebral and astrocytoma tissues

摘要 目的探讨过氧化物酶体增殖物激活受体(PPAR)γ在正常人脑组织和人脑星形细胞瘤组织中表达的差异。方法应用半定量RT-PCR法和免疫组织化学检测正常脑组织、不同分化级别人脑星形细胞瘤组织标本中PPARγ表达。结果PPARγmRNA和PPARγ蛋白在正常人脑组织、人脑星形细胞瘤中均可表达,且人脑星形细胞瘤中的表达显著高于正常脑组织(P<0.05),其中,Ⅰ~Ⅱ级组和Ⅲ~Ⅳ级组的表达均显著高于正常脑组织(P<0.05);Ⅲ~Ⅳ级组的表达显著高于Ⅰ~Ⅱ级组(P<0.05)。结论PPARγ在人脑星形细胞瘤和正常脑组织中均表达,与人脑星形细胞瘤的病理分级有关,可能参与人脑星形细胞瘤的发生、发展过程。 Objective To investigate the relationship between peroxisome proliferator- activated receptors γ (PPART) expression and clinicopathological grade of astrocytoma. Methods The expression of PPARγ, mRNA and PPARγ protein was detected in human normal cerebral tissues and astrocytoma Specimens by using RT-PCR semi-quantity and immunohistochemistry methods. Results The expression of PPARγ mRNA and PPARγ protein have been detected in normal cerebral tissues and human astrocytomas Specimens. The expression level of PPARγ mRNA and PPARγ protein in human astrocytomas was considerably higher than those in normal cerebral tissues (P〈0.05) ; and the expression level of PPARγ mRNA and PPARγ protein in astrocytomas of grade Ⅲ - Ⅳ was considerably higher than those in astrocytomas of grade Ⅰ - Ⅱ (P〈0.05). Conclusion The expression of PPARγ have been detected in human astrocytomas and normal cerebral tissues, it may be correlation to the pathological grades of human astrocytomas, it may be involved in the tumorigenesis and progression of human astrocytomas.
