摘要
目的研究榄香烯抗肿瘤的作用机制。方法用流式细胞仪和琼脂糖电泳检测DNA断裂,透射电镜观察超微结构变化。结果260μmol·L-1榄香烯孵育K562细胞6、12、24、48h后流式细胞仪检测到逐渐增强的凋亡峰,电泳发现明显的阶梯状条带,电镜观察到染色体聚集、凋亡小体。结论榄香烯能诱导K562细胞凋亡。
AIM To study the anticancer mechanisms of elemene. METHODS Endonucleolysis was assessed by agarose gel electrophoresis and flow cytometric methods. Morphological assessment of apoptosis was performed with electron microscope. RESULTS The K562 cells exposed to elemene 260 μmol·L-1after 6, 12, 24, 48 h showed a time-dependent increase of apoptotic peak. garose gel electrophoresis of DNA from cells treated with elemene revealed 'ladder' pattern. The marked morphological changes including condensed chromatin, nuclear fragmentation and apoptotic body were observed. CONCLUSION Our results indicate that elemene can induce apoptosis in K562 cells.
