川芎嗪对脊髓急性损伤大鼠神经营养因子表达的影响 被引量:8

Influence of tetramethylpyrazine on the expression of neurotrophic factors in rats with acute spinal cord injury

摘要 目的探讨川芎嗪(TMP)对大鼠脊髓急性损伤(ASCI)模型脊髓神经生长因子(NGF)、脑源性神经生长因子(BDNF)表达的影响及其对脊髓损伤后脊髓功能的保护作用。方法取SD大鼠随机分为正常对照组、假治疗组和川芎嗪组并给予相应处理。采用改良Allen’s法造模,BBB评分对实验大鼠脊髓功能进行行为学评分;SABC法检测造模后大鼠损伤段脊髓NGF和BDNF的表达。结果随时间延长,术后SD大鼠BBB评分逐渐升高,术后7 d、14 d和21 d川芎嗪组BBB评分值均较假治疗组高(P均<0.05);川芎嗪组术后3 d7、d和14 d时NGF、BDNF阳性细胞数表达均较假治疗组明显增加(P均<0.05)。结论川芎嗪对脊髓继发性损伤有保护作用,这种保护作用可能与其能促进损伤段脊髓NGF、BDNF的表达有关。 Objective It is to approach the influence of tetramethylpyrazine (TMP) on the expression of nerve growth factor (NGF) and brain derived neurotrophic factor (BDNF) in rat spinal tissues with acute spinal cord injury (ASCI), and it's neuroprotective effect after ASCI. Methods SD rats were randomly divided into Normal group, Pseudo-therapy group, TMP group and accepted corresponding disposal. The ASCI models were made according to Allen's weight drop method. Basso Beattie Bresnahan (BBB) scores were used to evaluate the spinal cord functional deficits. SABC immunohistochemistry method was used to detect the expressions of NGF and BDNF. Results Rats treated with TMP had better outcomes in BBB scores than Pseudo-therapy group at 7,14, 21d after ASCI respectively( P 〈 0.05) ; The number of NGF and BDNF positive cells in TMP group was significantly higher than in Pseudo-therapy group at 3, 7, 14d after ASCI (P 〈 0.05). Conclusion TMP has neuroprotective effects on the secondary injury of ASCI through regulate the expression of NGF and BDNF.
