摘要
以常用动物中药鸡内金为原料,用氢氧化钠为溶剂提取得到动物多糖(粘多糖)。碱提取法可以从组织中对粘多糖进行较完全的提取,但粘多糖分子有可能从断裂的一端被碱进一步降解,因此所用碱的浓度、碱解温度及时间都应优选。本实验研究了料液比,提取温度,提取时间和碱解浓度对提取率的影响。用响应曲面进行优化,最佳提取条件为:碱浓度1.5mol/L、提取温度45℃,提取时间50min,料液比1:25,此条件下提取率可达1.98%。
The mucopolysaccharides were abstracted by alkali from Endothelium corneum. The effects of the materials fluids ration, extraction temperature ,time, alkali additive on extraction rate have been described. The optimizing extraction conditions were obtained from the response surface methodology : the fluids materials ration was 1 : 25, extraction temperature was 45 ℃, extraction time was 50 min, and alkali additive was 1.5 mol/L. The optimizing extraction rate was 1.98%.
