大鼠主动脉粥样硬化斑块中Nogo-B的表达

摘要 目的检测大鼠主动脉粥样硬化斑块中Nogo-B的表达。方法选取40只Wistar大鼠,随机分为对照组和实验组,每组20只。实验组制成动脉粥样硬化模型,对照组同期正常饲养,8周后处死大鼠,取主动脉部位,采用Northen Blot和免疫组织化学技术检测Nogo-B mRNA和蛋白水平的表达。结果与对照组比较,实验组大鼠主动脉粥样硬化斑块中内皮细胞Nogo-B蛋白表达降低(P<0.05);Nogo-B1 mRNA表达显著下降40%,差异有统计学意义(P<0.01),Nogo-B2 mRNA表达轻度下降,但差异无统计学意义(P>0.05)。结论大鼠动脉粥样硬化斑块中Nogo-B1 mRNA和蛋白水平表达降低,推测Nogo-B1可能在动脉粥样硬化斑块形成过程中起保护作用。 Objective To investigate the expression of Nogo-B in the atherosclerotic plaque of rat's aorta. Methods Forty Wistar rats were equally divided into 2 groups: control group and experi- ment group. The rats of experiment group were used to form atherosclerotic models. Protein and mRNA expression levels of Nogo-B were measured with Northern Blot and immunohistochemistry respectively. Results Tissue immunohistochemical staining of aortic specimens indicated that Nogo-B took part in neovascularization and proliferation of smooth muscle. Compared with control group,the expression of Nogo-B in endothelial cells of plaques of aorta of rats in experiment group decreased. The mRNA expression levels of Nogo-B1 in atherosclerotic plaques decreased by 40 % compared with those in normal aorta, while the expression of Nogo-B2 changed slightly. Conclusion The expression of mRNA and protein of Nogo-B1 in atheroselerotic plaque is positively correlated to the formation of atherosclerosis and Nogo-B1 may have preventive effect against the pathological process of atherosclerosis.
