摘要
以phenomenex LUX Cellulose-1(250mm×4.6mm,3μm)色谱柱,以α-溴代苯基乙基酮为衍生试剂,在流动相为乙腈-0.5mol/L高氯酸钠(26:74),流速为0.4mL/min,检测波长为235nm的条件下采用柱前衍生法测定左旋肉碱(L-肉碱)中右旋肉碱(D-肉碱)的含量。结果表明对映异构体L-肉碱、D-肉碱衍生物分离度均大于2,D-肉碱的检测限为0.2μg/mL,在1~50μg/mL范围内线性关系良好,相关系数γ=0.9998。结论是方法灵敏度高、重现性好,可用于左旋肉碱中右旋肉碱含量的测定。
A HPLC method for the determination of D-camitine in L-camitine was developed in this paper. Samples were derivatized with (α-bromo) methyl phenyl ketone and analyzed on phenomenex LUX Cellulose-1 column (250mm×4.6mm,3μm)at 235 nm using acetonitrile-0,5mol/L sodium perchlomte (26:74) as the mobile phase at a flow rate of 0.4mL/min. Result showed that resolution for the optical isomer of camitine was larger than 2.0 and the linear range of D-camitine was 1-50μg/mL (γ=0.9998). The limit of detection (LOD) of D-camitine of this method was 0.2 μg/mL. The improved HPLC method was reliable and effective, which can be used for determination of D-camitine in L- camitine.
