摘要
[目的]选育富硒灵芝高产菌株并研究其液体发酵性能。[方法]应用添加Na2SeO3的培养基筛选富硒能力较强、菌体生物量及菌体多糖产量均较高的菌株,并研究其发酵性能。[结果]筛选到菌株Ganoderma lucidumZ-4-25,在液体培养基中添加浓度0.08%的Na2SeO3即能有效促进菌体生长,在浓度0.16%的Na2SeO3液体培养基中培养192 h,灵芝菌粉有机硒含量达到13 000μg/g菌体。在添加浓度0.08%的Na2SeO3条件下,10 L发酵罐发酵216 h,菌体生物量干重达到26.82 g/L,灵芝菌粉有机硒含量达到9 800μg/g菌体。[结论]该研究为富硒灵芝的液体发酵生产奠定了基础。
[Objective] The aim was to study the selective breeding and fermentation performance of Se-enriched Ganoderma lucidum high-yield strain.[Method] Se-enriched Ganoderma lucidum Z-4-25 was obtained using culture-medium with Na2SeO3,and its fermentation performance was studied.[Result] The mycelial biomass was increased obviously when cultivated in liquid medium with 0.08% Na2SeO3.The selenium content of mycelia was 13 000 μg/g,when cultivated for 192 h in liquid medium with 0.16% Na2SeO3.The dry weight of mycelia was 26.82 g/L,the selenium content of mycelia was 9 800 μg/g,when cultivated for 216 h in 10 L fermentor with 0.08% Na2SeO3.[Conclusion] The research will provide the basis for the liquid fermentation production of Se-enriched mycelia of Ganoderma lucidum.
