摘要
目的建立大鼠肾移植急性排斥反应(AR)模型,进行移植免疫机理的研究。方法正常雄性Wistar大鼠肾脏为正常对照组(control组);Wistar大鼠作为供受者建立同系基因对照组(sTX组);SD大鼠为供者,Wistar大鼠为受者建立同种异体基因移植组(aTX组);SD大鼠为供者,Wistar大鼠为受者,术后给予环孢素A(CsA)免疫抑制建立同种异体基因移植免疫抑制组(aTX+CsA组)。术后第3、5、7天分别切取移植肾,进行HE、Masson、PAS、PASM染色,光镜下观察病理改变,参照Banff 97诊断标准,进行AR严重程度的半定量评分。结果与control组相比较,术后第3、5、7天aTX组AR半定量评分明显升高(P<0.01);sTX组AR半定量评分升高,但无统计学差异(P>0.05);aTX+CsA组AR半定量评分也升高,但无统计学差异(P>0.05)。术后第3、57、天aTX组与sTX组相比较AR半定量评分明显升高(P<0.01);与aTX+CsA组相比较AR半定量评分明显升高(P<0.01)。aTX组中两两比较,第5、7天AR半定量评分与第3天相比较明显升高(P<0.05),第5天和第7天评分相比较无统计学差异(P>0.05)。sTX组和aTX+CsA组中术后第3、5、7天两两比较均无统计学差异(P>0.05)。结论Wistar-Wistar大鼠肾移植后,AR出现晚,排斥程度轻微,可以作为大鼠肾移植急性排斥模型的对照组;SD-Wistar大鼠肾移植后可出现明显的AR,因此,SD-Wistar大鼠移植模型可以作为大鼠肾移植急性排斥模型;CsA治疗可以明显减轻或抑制大鼠肾移植AR的发生。
Objective To establish the model of rat renal acute rejection and investigate the mechanism of transplantation immunology. Methods Normal Wistar male rat kidney were used as control. Wistar rat which received Wistar rat renal grafts were used as syngenegic group (sTX). Wistar rat which received SD rat renal grafts were used as allogenegic group (aTX). Wistar rat which received SD rat renal graft with CsA treatment were used as immunosuppression group (aTX + CsA). Grafts were harvested at day 3,5, 7 and performed H&E, Masson, PAS, PASM staining. AR semiquantitative scores were analyzed by using microscope according to the standard of Banff 97. Results Compared with the normal control, AR semiquantitative scores increased significantly in aTX group at the day 3,5,7 after transplantation( P 〈 0.01 ); the score increase a little bit in sTX group, but there is no significant difference compared with control group ( P 〉 0. 05). AR semiquantitative score increased significantly in aTX group at the day 3,5,7 after trans- plantation compared with sTX group (P 〈 0.01 ). The score increased significantly at the day 5, 7 after transplantation compared the scores at day 3 in aTX group ( P 〈 0.05). No significant different can be found between sTX group and aTX + CsA group at the day day 3, 5, 7 after transplantation. Conclusion Only slightly rejection can be found in sTX group, this group can be used as the control of rat renal AR;AR can be found in SD renal grafts,so SD to Wistar renal transplantation can be used for rat renal graft AR mod- el;CsA treatment can reduce and suppress renal graft AR.
