时辰对异氟烷催眠ED_(50)和LD_(50)的影响 被引量:5

摘要 目的观察不同时辰对小鼠腹腔注射异氟烷催眠半数有效量(ED50)和半数致死量(LD50)的影响。方法昆明种小鼠90只,雌雄不拘,随机分为ED50组和LD50组,每组再分为3个亚组。分别于8∶00、14∶00和20∶00腹腔注射异氟烷,应用序贯法实验,测定并计算各时间点的催眠ED50和LD50。结果异氟烷在14∶00的催眠ED50低于8∶00和20∶00(P<0.05,P<0.01),8∶00和20∶00的催眠ED50相似(P>0.05);异氟烷各时间点的LD50相似(P>0.05)。结论异氟烷的催眠ED50具有时辰依赖性。 Objective To investigate the effects of circadian rhythm on the median effective dose(ED50) and median lethal dose(LD50) induced by isoflurane.Methods 90 mice were randomized into ED50 group and LD50 group,with each group divided into 3 subgroups.Isoflurane was intraperitoneally injected at 8∶00,14∶00 and 20∶00,respectively.The hypnotic ED50 and LD50 at different time points were determined by up-down sequential allocation in mice. Results The hypnotic ED50 of isoflurane at 14∶00 was significantly lower than that at 8∶00 and 20∶00(P0.05,P0.01).The hypnotic ED50 of isoflurane at 8∶00 was similar to that at 20∶00(P0.05).The values of LD50 at each time point were similar(P0.05). Conclusion The hypnotic ED50 of isoflurane was circadian-dependent.
