摘要
目的建立测定犬血浆中豆腐果苷质量浓度的反相高效液相色谱(RP-HPKC)法。方法以0.05mol/LNaH2PO4缓冲液(调pH=3.5)-甲醇(77∶13)为流动相,犬血浆样品经乙腈沉淀蛋白萃取后,采用RP-HPLC法测定犬血浆中豆腐果苷的质量浓度。结果犬血浆中豆腐果苷质量浓度的线性范围为0.025~5.000μg/mL,平均相对回收率为88.16%~97.56%,日内和日间精密度的RSD均小于12.38%,豆腐果苷的检测限为10ng/mL。结论所建立的检测方法简便、快速、灵敏度高,适用于豆腐果苷的药代动力学研究。
Objective To establish a RP- HPLC method to detect the concentration of helicid in dog plasma. Methods The mobile phase consisted of 0.05 mol/L sodium dihydrogen phosphate (pH3.5)-methanol(77 : 13) was used. After pretreated by acetonitrile, the plasma samples were analyzed by RP - HPLC. Results The linear range of helicid was 0. 025 - 5. 000 μg/mL. And the recovery rate was 88.16% - 97.56%. Both the within-day and between-day RSD were less than 12.38%. The minimum detective limit was 10 ng/mL. Conclusion This RP- HPLC method is accurate, stable and sensitive, which can be applied to the pharmacokinetic study of helicid.
