摘要
对河南省3个山羊品种(槐山羊、伏牛白山羊、牛腿山羊)共计27个个体的线粒体D-loop区(mtDNAD-loop)全序列进行了PCR扩增和测序分析。检测到98个变异位点,19个单倍型,平均核苷酸差异数(K)为18.13,核苷酸多样度(Pi)为1.65%,单倍型多样度(Hd)为0.92;4种碱基A、T、C、G的频率分别为31.1%、28.9%、25.8%和14.2%;多态位点的突变类型以转换为主;构建的NJ进化树分为3枝,槐山羊、伏牛白山羊、角羊骨羊和部分牛腿山羊聚为一枝,部分牛腿山羊单独聚为一枝,作为外群的捻角山羊单独为一枝。河南省3个山羊品种遗传多样性丰富,支持山羊品种的多起源说。
The complete mitochondrial control regions of 27 goats from 3 domestic breeds including Huai goat,Niutui goat and Funiu white goat of Henan province were cloned and sequenced.The results showed that a total of 98 variations and 19 haplotypes were detected.The average number of nucleotide differences( K ),nucleotide diversity( Pi )and haplotype diversity( Hd ) were 18.13,1.65% and 0.92,respectively.The frequencies for A,T,C and G bases were 31.1%,28.9%,25.8% and 14.2%.Among the 98 variations,there were most of transitions.The NJ tree showed three branches,in which breeds of Huai goats,Funiu white goats and some of the Niutui goats belong to one branch,the other of Niutui goats belong to the second branch and Capra falconeri belong to the branch of outgroup.These data indicate that the genetic diversity of three domestic goat breed in Henan province was abundant,supporting multi-origin theory of goat breed.
