摘要
[目的]为苹果锈果类病毒结构域功能的关系、复制机制、变异与进化的研究提供理论依据。[方法]应用生物信息学的方法,得到一系列苹果锈果类病毒的RNA序列,对比其核苷酸数目、碱基组成比例,并进行了多重系列对比及二级结构预测。[结果]来自不同地区的苹果锈果类病毒RNA和核苷酸数目分布在329~336bp。其中核苷酸数为330bp的种类占64.71%,其A的分布范围19.39%~20.61%,T的分布范围在18.79%~20.91%,G的分布范围在29.70%~30.61%,C的分布范围在30.00%~30.91%。用RNAstructure软件进行苹果锈果类病毒的二级结构预测,随着稳定能量级的逐渐增加,其碱基配对区逐渐减小,而茎环区却逐渐增加。[结论]RNA是生物生命活动不可或缺的重要生物大分子,其二级结构比一级结构具有更大的保守性。
[Objective] The theoretical basis of the study on the relationship of the structure function,the replication mechanism,the variation and evolution of apple rust fruit-like virus was provided.[Method] A series of RNA sequences of apple rust fruit-like virus were produced with the bioinformatics methods.The number of nucleotide and the proportion of base composition were compared and the comparison of multiple series and prediction of its secondary structure were conducted.[Results]The nucleotide number of RNA of the apple rust fruit-like virus from different regions was found in the range of 329-336 bp,of which types,the rate of the nucleotide number=330 bp accounted for 64.71%,which distribution of A was 19.39%-20.61%;T,18.79%-20.91%;G,29.70%-30.61% and C,30.00%-30.91%.The prediction result of its secondary structure with RNAstructure software indicated that the zone of its base pairing was gradually reduced but the stem-loop region was gradually increased with the gradual increment of stable energy level.[Conclusion] RNA is an indispensable and important biological macromolecules in biological life activity and its secondary structure is more conservative than the first structure.
