BALB/c小鼠子宫内膜异位症模型的建立及其形态学的动态观察 被引量:14

摘要 目的研究BALB/c小鼠子宫内膜异位症模型的建立及异位病灶形态学的动态变化规律。方法采用组织块腹腔注射法对110只BALB/c小鼠行建模手术,在术后第1、2、3、4、5、6、9、12、15、18、21天的11个观察日里,每次观察10只小鼠,了解病灶的生长情况并进行组织学的光镜和电镜观察。结果建模术后第2天开始出现病灶,第6天病灶都是红色,第9天开始出现粘连带,第15天后大部分病灶呈透亮的小囊泡。第4天的建模成功率为90%(9/10),与第21天的建模成功率无显著性差异(P=1.000)。第4天异位灶的重量(51.5±4.7)mg与第21天(68.9±7.2)mg无显著性差异(P>0.05)。光镜下见子宫内膜上皮细胞和间质细胞,电镜下见细胞间完整的紧密连接,表面微绒毛。结论组织块腹腔注射法可成功构建BALB/c小鼠子宫内膜异位症模型,术后不同时间内异位灶的形态学呈现特定的变化规律。 Objective To establish a BALB/c endometriosis model and observe the dynamic morphological changing pattern of the ectopic focus.Methods By using intraperitoneal infection of endometrial tissues,BALB/c model of endometriosis was established in 110 mice.The mice were then randomly divided into 11 months to be killed at the 1st,2nd,3rd,4th,5th,6th,9th,12th,15th,18th and 21st day after the operation(10 mice in each group)to observe the development and histology of the ectopic focus by light and electron microscopy.Results The ectopic focus began to appear on the 2nd day,presenting red color on the 6th day.Then adhesion took place on the 9th day and all the lesions evolved into white or transparent cysts on the 15th day.The success rate of modeling after the 4th day was 90%(9/10),showing no significant difference with that in the following days(P〉0.05).The weight of ectopic focus on the 4th day was(51.5±4.7)mg,which was not significantly different from that weighed(68.9±7.2)mg on the 21st day(P=1.000).Under a light microscope,endometrial epithelial cells and stromal cells were seen,and they showned close linked with each other with micro tomenta on the surface under an electron microscope.Conclusions We established BALB/c endometriosis models by using intraperitoneal infection of endometrial tissues.The morphology of the ectopic focus has a particular changing pattern at different periods of time after the operation.
