摘要
为了筛选出卷叶贝母愈伤组织快速增殖的最佳培养条件,从不同激素配比、培养温度、光照强度与时间等方面筛选出卷叶贝母愈伤组织快速增殖的最佳培养条件,并利用紫外分光光度法测定其总生物碱含量。结果显示,愈伤组织增殖的最佳培养基为MS+6-BA 2.0 mg.L-1+NAA 0.5 mg.L-1,最适温度为20℃,最适光照时间12±2 h.d-1,在此培养条件下卷叶贝母愈伤组织培养40 d可增殖3倍以上。
To screen the optimal culture conditions for fast proliferation of Fritillaria cirrhosa D. Don callus, the factors including hormone combination ,culture temperature,light intensity and inoculum size were analyzed. The total alkaloid content was measured by UV spectrophotomctry. The results showed that the best culture medium for callus proliferation was Ms+6.BA2.0mg·L^-1+NAA0.5mg·L^-1;the optimum temperature was 20 ℃, the optimum illumination time was 12 ± 2 h· d^-1 Under such culture condition, Fritillaria cirrhosa D. Don callus could proliferate more than 3 times after cultured for 40 d.
