摘要
目的:建立以高效液相色谱法(HPLG)测定人血浆中异烟肼浓度的方法。方法:色谱柱为CLC-ODS,以磷酸二氢钾溶液∶乙腈=98∶2(用氨水调节pH约为5.6)为流动相,检测波长为270nm。结果:异烟肼的线性范围为0.064~39.74μg/mL,平均回收率为98.82%。结论:本法专属性高,样品处理方法简单,可用于测定人血浆中异烟肼的浓度。
Objective:To determine the concentration of plsama isoniazid by RP-HPLC.Methods:The HPLC system consisted of CLC-ODS column,0.05 mol/L KH2PO4-acetonitrile(98:2,adjusting pH to 5.6)as the mobile phase,the flow rate was 1.0 mL/min,the column temperature was 25℃ with the detection wavelength at 70 nm.Results:The linear ranges of isoniazid was 0.064~39.74 μg/mL(r=0.9932).The extraction recovery rate was 98.82%.Conclusion:The assay in this study is sensitive,simple and rapid.It could be used to determine the concentration of isoniazid in human plasma.
