以DTNB为电子媒介检测农产品中甲拌磷残留的生物传感法的建立 被引量:3

摘要 目的:建立一种检测有机磷残留的生物传感法。方法:基于壳聚糖膜,用戊二醛交联法固定胆碱氧化酶制备胆碱传感器。测试时在反应液中加入5,5-二硫代双(2-硝基苯甲酸)(DTNB),增大传感器对硫代胆碱的响应信号,建立测试有机磷的反应体系。结果:用该传感方法检测有机磷农药甲拌磷,与分光光度法比较,测试结果相近,无显著差异(p>0.01)。检测甲拌磷线性范围0.78~12.5mg/L,检测限为0.02mg/L。结论:实验建立的检测有机磷农药的传感方法稳定、简便、快速,可望改进为成熟的机磷农药现场快速检测技术,从而为农产品监测提供有力的手段。 Object:To develop a biosensor method for orgnaophosphorus residue determination.Methods:Choline oxidase was immobilized on chitosan membranes by glutaric dialdehyde method and the change in oxygen consumption upon orgnaophosphorus introduction was measured.5,5'-Dithiobis(2-nitrobenzoic acid)(DTNB) was added to increase the response currents.Results:Compared with spectrophotometric method,there was no statistically significant deviation and the linear range for phorate determination was 0.78~12.5 mg/L,the detection limit 0.02 mg/L.Conclusion:This method is stable,convenient,rapid,and can be developed into a real time method for for orgnaophosphorus determination in vegetables and other agricultural products.
