摘要
目的观察注射外源性脂联素对ob/ob小鼠2型糖尿病心肌病微血管病变的影响。方法20只ob/ob小鼠高热量饮食诱导2型糖尿病,随机分为糖尿病心肌病组和重组脂联素干预组,干预组给予重组脂联素10μg/(kg.d)腹腔注射,持续8周;以10只C57/BL野生型小鼠作对照。测定血糖、血脂,血清脂联素水平。心肌切片做HE染色,AB-PAS染色,同时送电镜观察。结果与对照组比较,糖尿病心肌病组小鼠血糖、血脂水平明显增高(P<0.01),血清脂联素水平明显降低(P<0.05);与糖尿病心肌病组比较,重组脂联素组血糖、血脂水平明显降低(P<0.01),血清脂联素水平明显增高(P<0.05);与对照组比较,糖尿病心肌病组微血管变重,糖原沉积明显,重组脂联素组微血管病变轻,糖原沉积不明显;血清脂联素水平与血糖、血脂负相关。结论脂联素与糖尿病微血管病变相关,注射外源性重组脂联素能够降低2型糖尿病小鼠的血糖、血脂,减少心肌微血管的糖原沉积,保护血管内皮功能,从而减缓糖尿病心肌病的发展进程。
Objective To observe the effect of exogenous adiponectin on diabetes,myocarditis,microvascular disease of ob/ob mice.Methods Ob/ob mice were fed with high fat diet to induce type 2 diabetes(fasting plasma glucose 13.8mmol/L),randomly divided into disease group and recombinant adiponectin intervention group,the recombinant adiponectin intervention group were intraperitoneal injected with recombinant adiponectin 10 μg/(kg·d)for 8 weeks;compared with type C57/BL wild mice.The level of glucose,lipid,serum adiponectin were tested at the end of the experiment.Its myocardial slice was made by HE colouration,AB-PAS colouration,at the same time,the sample was sent to electrical microscope observation.Results Compared with sample group,the glucose,lipid level of mice in diabetes group was obviously higher(P〈0.01),the level of serum adiponectin was sharply lower than that of the sample group(P〈0.01);Compared with diabetes group,the glucose and lipid levels of recombi-nant adiponectin intervention group were obviously lower(P〈0.01),the level of serum adiponectin was sharply higher(P〈0.01).Compared with sample group,the degree of microvascular disease of mices in diabetes group went heavier,clearer glycogen deposition could be seen.The degree of microvascular disease in adiponectin intervention group was relatively lighter,and the glycogen deposition in this group was not obvious.There was a negative correlation relationship between serum adiponectin and glucose,lipid.Conclusion There are definitely relationships between adiponectin level and diabetes,myocarditis,microvascular disease.Exogenous adiponectin injection could decrease the glucose,lipid and the glycogen deposition of myocardial microvascular of type 2 diabetes mice;it has a function of vascular endothelial protection,so it can slow down the speed of the development process of diabetes and myocarditis.
