摘要
通过正交试验得出Arazyme蛋白酶酶解地鳖虫的最佳工艺条件为加酶量300U,酶解时间5h,料水比1:20。并分析了其酶解液对羟自由基(·OH)、超氧阴离子自由基(O2-·)、二苯代苦味酰基自由基(DPPH)的清除作用以及对亚油酸自氧化反应的抑制作用。结果表明:地鳖虫酶解液对·OH、O2-·、DPPH三种自由基的最高清除率分别为75.15%(浓度为50g/L)、31.72%(浓度为2.5g/L)、74.88%(浓度为80mg/L);在肽含量为2.0mg时,对亚油酸自氧化反应的抑制率为95%。
The optimum conditions of enzymatic hydrolysis by Arazyme protease of Eupoliphaga Sinensis Walker were enayme concentration 300 U,time of hydrolysis 5 h,and solid-to-liquid ratio 1:20.The free radicals scavenging activity on·OH,O2^-·,DPPH and linoleic acid oxidation inhibition of hydrolysate was invistgated.The result showed that the highest scavenging activityof·OH,O2^-·and DPPH was 75.15 %(concentration 50 g/L),31.72 %(concentration 2.5 g/L),74.88 %(concentration 80 mg/L).The linoleic acid oxidation inhibition was 95 % (concentration of peptide was 2 mg).
