摘要
以乳酸链球菌素效价和菌体生物量为检测指标,通过单因素试验研究碳源、氮源、磷酸盐、金属离子和发酵条件对乳酸链球菌肽菌株(Lactococcu lactis subsp.lactics)W4摇瓶发酵产生乳酸链球菌素的影响。结果表明:葡萄糖是W4菌株发酵的最佳碳源,酵母膏为最佳氮源,K2HPO4、Fe2+、Mg2+对其发酵有促进作用,而Cu2+有强抑制作用;初始pH6.5、2%接种量、35℃下振荡培养有利于W4菌株生长和乳酸链球菌素的产生。
The effects of medium composition and fermentation condition on Lactococcu lactis subsp.lactics W4 producing nisin were studied through single factor experiment.Nisin titer and biomass were used as objective product.The results showed that glucose was optimum carbon sources,yeast extract was optimum nitrogen sources.The fermentation of the strain W4 was promoted by K2HPO4,Fe^2+ and Mg2+.The fermentation was restrained by Cu^2+.Optimal original pH was 6.5,filled volume was 2%.Shaking culture at 35℃.
