摘要
本文运用溶析结晶原理研究了第三代半合成青霉素美洛西林钠结晶工艺,包括反应溶剂、溶析剂、温度、搅拌速率、pH值等因素对产品质量的影响及工艺条件的优化。结果显示,丙酮作为反应溶剂,溶析剂为异丙醇,得到美洛西林钠收率95.0%,纯度99.0%(HPLC)。该工艺过程具有操作简便,反应效率高,溶剂易回收套用,杂质易去除,收率高,产品色泽好,纯度高,溶解性好等优点。
The solvent-out crystallization process of mezlocillin sodium for injection was studied. Varied factors which affected the crystallization process were investigated and the crystallinic conditions were optimized. It includes various kinds of reaction solvent, the solvent-out solvents, temperature, stirring rate, value of pH and so on. The process was carried out using acetone as the reaction solvent and isopropyl alcohol as solvent-out solvent. The yield was 95.0% with high purity of 99.0% (HPLC). The method of preparation has the advantages of simple operation, higher efficiency of reaction, easily solvent recycling, removal of impurity from product, good yield and color, purer product and better solubility.
