益气养阴、清利活血中药对微小病变肾病大鼠肾组织TGF-β表达的影响 被引量:6

摘要 目的:观察益气养阴、清利活血中药对阿霉素诱导的微小病变肾病大鼠肾组织转化生长因子-β(TGF-β)表达的影响,探讨其肾脏保护作用机制。方法:实验动物随机分为6组:正常组,模型组,阳性对照组,中药高、中、低剂量组。用表柔比星(阿霉素)尾静脉注射法(6mg/kg)复制微小病变肾病大鼠模型,分别给予不同剂量益气养阴、清利活血中药液、强的松混悬液。测定大鼠24小时尿蛋白定量、血清白蛋白(ALB)、总胆固醇(TC)、甘油三酯(TG)、尿素氮(BUN)、肌酐(SCr)及肾组织TGF-β(ELISA法)的含量,并电镜下观察肾脏组织形态学变化。结果:与正常组比较,模型组大鼠肾组织TGF-β含量增高(P<0.01);益气养阴、清利活血中药可减少微小病变肾病大鼠尿蛋白的排泄量,改善血清生化指标,降低肾组织TGF-β含量(P<0.05~0.01),修复肾小球上皮细胞足突的结构。结论:益气养阴、清利活血中药可抑制微小病变肾病大鼠增高的TGF-β表达,从而减轻肾脏病理损害。 Objective:To observe the effect of tonifying qi nourishing yin,clearing and promoting blood circulationChinese herbs on TGF-β expression in kidney of rats with minimal nephritic syndrome,to explore the mechanism of protecting kidney.Methods:48 rats were divided into normal,model,positive,Chinese herbs high,middle and low dose groups,models with minimal nephritic syndrome were made by ADR injected through caudal vein.Each group was given corresponding medicine for 4 weeks.The 24h urinary protein,serum ALB,TC,TG,BUN and SCr were determined respectively,kidney TGF-β content was detected by ELISA,and the morphologic changes of kidney were observed by electron microscope.Results:To compare with normal group,the 24h urinary protein,serum TC,TG,BUN and SCr were increased,ALB was decreased,kidney TGF-β content was raised;tonifying qi nourishing yin,clearing and promoting blood circulation Chinese herbs could reduce the 24h urinary protein,lower serum TC,TG,BUN and SCr level and elevate ALB level,lower kidney TGF-β content,repair the injured podocytic process structure of glomerular epithelial cells.Conclusion:Tonifying qi nourishing yin,clearing and promoting blood circulation Chinese herbs can inhibit the over-expression of TGF-β in minimal nephritic syndrome rats to abate the pathologic injury of kidney.
