摘要
目的研究18β-甘草酸对大黄酸在大鼠体内药物动力学的影响。方法大鼠分别灌服大黄酸和甘草酸+大黄酸,高效液相色谱法测定大黄酸的血药浓度。色谱柱为Zorbox SB C18(150mm×4.6mm,5μm),流动相为甲醇-水-冰醋酸(77∶22∶1);检测波长为428nm;流速为1.0mL·min-1;柱温为室温。血药浓度数据采用3P97药物动力学软件进行分析。结果大黄酸血药浓度在0.1~15μg.mL-1范围线性关系良好。大鼠灌服大黄酸和甘草酸+大黄酸后,大黄酸血浆浓度-时间曲线均符合二房室模型。灌服大黄酸组和甘草酸+大黄酸组相比较,两者的AUC、Cmax,均存在显著性差异(P<0.05)。结论 18β-甘草酸可影响大黄酸在大鼠体内的药物动力学过程,使大黄酸在大鼠体内的血药浓度和生物利用度降低。
Objective To study the effect of 18β-glycyrrhizic acid on pharmacokinetics of rhein in rats.Methods Sprague Dawley rats were given gastric gavage of rhein and glycyrrhizic acid+rhein respectively.The plasma concentration of rhein was measured by high performance liquid chromatography.HPLC method was established with Zorbox SB C18 column(150 mm×4.6 mm,5μm),the mobile phase was methanol-water-acetic acid(77∶22∶1),the flow rate was 1.0 mL·min^-1 and detection wavelength was 428 nm.The pharmacokinetic parameters were determined by analyzing the plasma concentration-time data with software 3P97.Results A good linear relationship of rhein was obtained in the range of 0.1~15μg·mL^-1.The concentration-time curves of rhein were shown as two-compartment models in rats.Significant difference was found between the two groups.There existed significant differences in AUC and Cmax between rhein group and glycyrrhizic acid+rhein group(P〈0.05).Conclusion The concentration and the bioavailability of rhein in rats can be decreased by 18β-glycyrrhizic acid.
