摘要
目的探讨不同价态及剂量砷染毒对大鼠血清丙酮酸激酶(pyruvate kinase,PK)的影响,为阐明砷毒作用机制提供依据。方法 Wistar大鼠70只,采用喂饲法染毒,暴露于不同价态无机砷。动物随机分成7组,每组10只,雌雄各半:即正常对照组(C);亚砷酸钠低(AL)、中(AM)、高(AH)剂量组;砷酸钠低(BL)、中(BM)、高(BH)剂量组。大鼠饲养至第3个月末,用颈椎脱臼法处死。应用酶联免疫分析法(ELISA)测定大鼠在不同价态及剂量砷染毒后血清PK活性变化。结果血清PK活性在亚砷酸钠和砷酸钠高剂量组中的比较差异有统计学意义(P<0.05),在亚砷酸钠和砷酸钠中剂量组与低剂量组比较时差异均无统计学意义(P均>0.05),在亚砷酸钠和砷酸钠与对照组比较时差异均有统计学意义(P均<0.05)。结论不同价态高剂量无机砷染毒,可导致PK活性降低,并且机制不完全相同。PK活性的改变可能是影响砷代谢及其毒作用的重要环节,低剂量染砷未见PK活性的改变。
Objective To study the influence on serum pyruvate kinase(PK) activity between arsenic in different valence state and doses for the mechanism of toxic action.Methods Seventy wistar rats was treated with drinking water containing different valence state and doses of arsenic,both of female and male was 35;and rats were randomly divided into 7 group,including control group(C),sodium arsenite low dose group(AL),middle dose group(AM),high dose group(AH);sodium arsenate low dose group(BL),middle dose group(BM),high dose group(BH).At the end of the 3 months the rats were executed by cervical dislocation.The serum pyruvate kinase activity was detected by ELISA in different valence state and doses.Results Comparing the activity of serum pyruvate kinase between sodium arsenate and sodium arsenite,the differences of high dose group was statistically significant between different valence state arsenic(P0.05).The middle dose and low dose group was not statistically significant(P0.05).The differences between every dose group and control group were statistically significant(P0.05).Conclusion High dose arsenic in different valence state may decrease the activity of serum pyruvate kinase,and the mechanism of toxic action was not completely identical.The alteration of pyruvate kinase activity was likely to be a key link of metabolism and toxic action of arsenic,and which would exert an important influence on it.Low dose arsenic unchanged the activity of pyruvate kinase.
