不同介质对线虫卵孵化实验效果的影响

摘要 【目的】用饱和盐水与饱和蔗糖溶液收集绵羊消化道线虫卵,在相同的条件下进行虫卵孵化实验,探究不同的介质对虫卵孵化试验效果的影响。【方法】按照世界兽医寄生虫学促进学会(WAAVP)推荐的虫卵孵化试验(egg hatch test,EHT),将经饱和盐水和饱和蔗糖溶液收集的线虫虫卵,用24孔培养板分4组进行重复实验,观察虫卵的孵育情况,并统计分析其差异性。【结果】经统计分析,用饱和蔗糖溶液收集的虫卵其虫卵孵育率较饱和盐水高,其差异极其显著(P<0.01),经2种介质收集的虫卵均可良好发育。【结论】经饱和盐水与饱和蔗糖溶液收集的线虫卵用于虫卵孵化试验的结果存在差异,但实验结果证实2种介质收集的虫卵均可用于虫卵孵化试验及抗药性的检测,但在具体的虫卵发育实验应用过程中应注意收集方法的统一性。 [ Objective]The nematodes eggs were collected by saturated saline and saturated sucrose, then egg hatch test was conducted under the same conditions to probe the effect of different mediator on the result of egg hatch test. [Method]According to the method(egg hatch test) recommended by the World Associatia the Advancement of Veterinary Parasitology (WAAVP), the nematodes eggs collected with saturated saline and saturated sucrose solution were tested repeatedly in four groups using 24 - hole culture board. The egg hatching development was observed and the variability was statistically analyzed. [Result]The development rate of eggs collected by saturated sucrose was higher than saturated saline , the variance was extremely significant( P 〈 0.01 ) . Eggs collected by the two mediators can both develop well. [ Conclusion] The eggs collected by saturated saline and saturated sucrose exist variance in the result of egg hatch test, but they can be used on egg hatch test and the resistance detecting, but the uniformity of the collecting method should be noted in practical application.
