甜瓜杂交种纯度电泳鉴定方法研究 被引量:6

摘要 对甜瓜杂交种8501和8601及其母本的几种种子蛋白和几种同工酶进行了电泳分析。结果表明.用蒸馏水、5%NaCl、70%乙醇、6%尿素和0.05mol/LHCl提取的各组分种子蛋白的电泳图谱在母本和杂种间均无明显差异。种子和6d苗龄子叶的谷氨酸脱氢酶同工酶、超氧化物歧化酶同工酶、酸性磷酸酯酶同工酶、过氧化物酶同工酶以及种子的酯酶同工酶酶谱在母本和杂种间也未见差异,但10d苗龄子叶的过氧化物酶同工酶酶谱在母本和2个杂种间均表现出明显差异,6~12d苗龄子叶的酯酶同工酶酶谱在母本和8501(F1)间也表现明显差异,且酯酶同工酶的这一结果不受实验所用的4种提取液的影响。利用过氧化物酶同工酶和酯酶同工酶对甜瓜杂种8501和8601进行了室内和田间检测结果对比,符合率达97%以上。 The seed purity was analysed by seed proteins and isozyme electrophoresis in two melonhybrids 8501, 8601 and their common fcmale parent as control. The results indicated that there were nosignificant difference between the hybrids and the female parcnt in clcctrophoretogram of seed proteinsextracted with H2O, 5% NaCl, 7O% ethanol, 6% urea and 0. 05 mol/L HCI separately, and nosignificant difference isozymegrams of GDH, SOD, ASP and POD from seeds and cotyledons of 6-day-oldseedlings. However, there were significant difference in POD in the cotylcdons of 10-day-old seedlingbetween hybrids and parent, and of EST ln the cotylcdons of 6 to 12-day-old seedlings between 8501 andthe parent. It was found that isozymegrams of EST was idcntical extracted with four different ways inlaboratory. The result for testing hybrid purity of 8501 and 8601 using POD and EST was compared withfield test result, the corresponding ra1e was over 97 %.
