摘要
为了提高黄檗组织培养的愈伤组织诱导率,研究了植物激素、基本培养基、外植体、蔗糖质量浓度、pH值和光照等因子对黄檗愈伤组织诱导和生长的影响。结果表明,以叶片为外植体,MS为基本培养基,培养基中添加BA1.5 mg/L和2,4-D 2.0 mg/L,培养基中蔗糖质量浓度20 g/L,pH值5.8,在光照强度40μmol.m-2s-1条件下培养最有利于黄檗叶片愈伤组织的诱导。在此条件下,愈伤组织诱导率最高可达100%。
In order to enhance callus induction rate,effects of several factors on induction and growth of callus in Phellodendron amurense were researched,for example,plant hormones,basic media,explants,sucrose mass concentrations,pH values and light intensity.The results showed that taking leaves as explants and MS as basic medium,the medium supplemented with BA 1.5 mg/L,2,4-D 2.0 mg/L and sucrose 20 g/L was of benefit to callus induction in P.amurense,cultivating under the conditions of pH 5.8 and light intensity 40 μmol·m-2s-1.Callus induction rate was up to 100% under the conditions.
