摘要
[目的]探索白芨种子萌发的适宜培养条件。[方法]以MS为基本培养基,分别考察灭菌剂浓度、基本培养基种类、光照周期、蔗糖浓度、活性炭浓度对白芨种子萌发的影响。[结果]白芨种子萌发的适宜培养条件为:采用1%的次氯酸钠灭菌,1/2MS培养基,2%蔗糖,0.1%活性炭,光周期10-12 h/d,在该培养条件下7 d后白芨种子萌发率可达80%以上。[结论]为白芨组织培养快速繁殖体系建立提供理论依据。
[Objective]The aim was to study the suitable culture conditions of Bletilla striata seed germination.[Method] The factors which effected the seed germination were studied such as types of minimal medium,illumination periodicity,concentration of sterilizing agent,sucrose and activated carbon etc.based on the MS minimal medium.[Result] The suitable culture conditions of Bletilla striata seed germination were as follows: 1% sodium hypochlorite for the seeds sterilization,1/2MS nutrient medium with 2% sucrose and 0.1% activated carbon,illumination periodicity 10-12 h/d.The seeds germination rate was above 80% after 7 days under above culture condition.[Conclusion] The study provided theoretical basis for the establishment of Bletilla striata rapid propagation system.
