灯盏花素脂质体前体与灯盏花素片抗血栓作用比较研究 被引量:1

Comparative investigation of the effect of breviscapine tablet and breviscapine proliposome on thrombosis of rats

摘要 目的我们前期研究结果表明灯盏花素脂质体前体(PS)减少大脑中动脉栓塞引起的脑梗死面积,改善脑缺血再灌注损伤作用明显优于灯盏花素片(BreT),本文对PS和BreT对血栓形成和溶栓作用进行比较研究。方法线栓法制作大脑中动脉阻塞脑缺血-再灌注损伤模型,经2,3,5-氯化三苯基四氮唑(TTC)染色,检测大鼠脑梗死面积;建立大鼠动-静脉旁路血栓模型、下腔静脉血栓模型、颈总动脉血栓溶栓模型检测血栓湿重;毛细玻管法检测大鼠血小板数目及小鼠凝血时间。结果 PS及BreT均显著减少脑梗死面积(P<0.05),PS作用强于同等剂量的BreT(P<0.05)。PS及BreT组均不同程度降低大鼠动-静脉旁路、下腔静脉及颈总动脉血栓湿重。血栓形成抑制率及溶栓率,PS作用强于同等剂量的BreT。PS及BreT显著延长小鼠凝血时间(P<0.01),PS与同等剂量的BreT相比具有显著性差异(P<0.01)。各组给药前后血小板数量无显著性差异(P>0.05)。结论灯盏花素脂质体前体抑制血栓形成和溶栓作用明显优于灯盏花素片,该作用与其增强抗脑缺血-再灌注损伤作用有关。 Objective We have previously found that the novel preparation breviscapine proliposome (PS) produced stronger effects than breviscapine proliposome(Bre T) in amelioration of cerebral ischemia-reperfusion injury. To study the potential role, a comparative investigation between PS and Bre T on thrombosis and thrombolysis was conducted. Methods The middle cerebral artery occlusion model was made by intraluminal filament technique, the cerebral infarct area was measured by TTC-staining. Carotid arterio-venous bypass thrombus model and vena cava inferior thrombus model were established to investigate antithrombotic effect of PS, carotid artery thrombus was established by exposure to FeCl3 to investigate thrombolytic effect of PS, wet weight of thrombus was measured. Clotting time was determined by capillary method, platelet number was counted before and after PS or Bre T administration, with Bre T as a positive control. Results PS decreased cerebral ischemia-reperfusion infarct area and had higher protective effect compared with Bre T ( P 0.05). The wet weight of thrombus in PS group and Bre T group was decreased respectively, the inhibition ratio of thrombus and thrombolytic was higher in PS group than in Bre T group. PS and Bre T prolonged coagulation time (P0.01), but stronger for PS compared with Bre T (P 0.01). There was no significant difference for platelet number before and after PS or Bre T administration. Conclusion The effects of breviscapine proliposome on inhibition of thrombosis and promotion of thrombolysis are significantly stronger than Bre T in cerebral ischemia-reperfusion injury induced by middle cerebral artery occlusion in rat.
