摘要
目的探讨缺血性脑卒中患者血小板活化标记物血小板糖蛋白Ⅵ(glycoprotein,GPⅥ)、P-选择素(CD62P)的表达水平及葛根素治疗对其的影响。方法收集2009年1-6月佛山市第三人民医院首发的缺血性脑卒中住院患者50例和同期体检的正常对照50例作为研究对象;采用流式细胞仪(FCM)检测两组GPⅥ和CD62P的表达水平并观察患者葛根素治疗前后GPⅥ和CD62P的表达变化;应用双抗体夹心ELISA法检测两组血浆中可溶性GPⅥ水平;应用直线相关分析统计血小板膜GPⅥ与CD62P、血小板膜GPⅥ与可溶性GPⅥ之间的相关性。结果缺血性脑卒中患者(病例组)血小板膜GPⅥ和CD62P表达增加,并且明显高于正常对照组(P<0.01);葛根素治疗后血小板膜GPⅥ、CD62P表达有明显下降(P<0.05),但仍高于正常对照组(P<0.05);缺血性脑卒中患者血小板膜GPⅥ与CD62P之间呈显著直线正相关(r=0.784,P<0.01);病例组可溶性GPⅥ水平明显低于正常对照组(P<0.01);血小板膜GPⅥ与血浆可溶性GPⅥ之间呈直线负相关(r=-0.828,P<0.01)。结论 GPⅥ和CD62P均可作为缺血性脑卒中血小板活化的标志物,而且二者具有同样的敏感性;GPⅥ、CD62P检测对于指导用药有临床价值;血浆可溶性GPⅥ亦可作为血小板活化的评估指标。
Objective To study the effect of puerarin treatment on expression of platelet activation biomarkers,glycoprotein Ⅵ (GPⅥ) and CD62P, in patients with ischemic cerebral stroke. Methods Expression of GP Ⅵ and CD62P was detected by flow eytometry (FCM) before and after puerarin treatment in 50 isehemic cerebral stroke patients from Jan. to Jun. 2009 in Foshan Third People' s Hospital. Another 50 health individuals served as normal control. Soluble GPⅥ( sGPⅥ ) in plasma of patients with ischemic cerebral stroke was assayed with double antibody sandwich ELISA. Correlation between expressions of platelet GPⅥ and CD62P, and between expressions of platelet and sGPⅥ was observed by linear correlation analysis. Results The expression level of platelet GP Ⅵ and CD62P was significantly higher in patients with ischemic cerebral stroke than in normal control group ( P 〈 0.01 ) and was significantly lower after than before treatment with puerarin( P 〈 0.05 ), but still higher than in normal control group ( P 〈 0.05 ). A significant positive linear correlation was observed between expressions of platelet GPⅥ and CD62P (r = 0. 784, P 〈 0.01 ). The sGPⅥ level was significantly lower in patients with ischemic cerebral stroke than in normal control group(P 〈0.01 ). A significant negative linear correlation was found between expressions of platelet GPⅥ and sGPⅥ (r = - 0. 828, P 〈 0.01 ). Conclusion Both GPⅥ and CD62P can be used as biomarkers of platelet activation in patients with isehemic cerebral stroke with a similar sensitivity. Detection of GPⅥ and CD62P helps rational use of drugs in clinical practice. Plasma sGPⅥ can also be used as a parameter for the assessment of activated platelets.
