南方红豆杉离体胚培养诱导不定芽研究 被引量:10

Regeneration of Taxus chinensis var. mairei from adventitious bud formation using an in vitro embryo culture

摘要 以南方红豆杉Taxus chinensis var.mairei成熟胚为外植体,筛选出外植体灭菌途径,研究了基本培养基、生长调节剂等因子对其离体胚生长的影响。结果表明:最优灭菌处理为25.0g·L-1的次氯酸钠(NaClO)溶液真空灭菌10min,其污染率仅为6%,出苗率可达86%;木本植物培养剂(WPM)为胚培养的最适基本培养基;细胞激动素(KT)0.1mg·L-1和玉米素(ZT)1.0mg·L-1最有效于不定芽的诱导,不定芽为丛生芽;6-苄氨基腺嘌呤(6-BA)和异戊烯基腺嘌呤(2iP)诱导不定芽较少;噻苯隆(TDZ)不能诱导不定芽;以萌动种胚为外植体对不定芽的诱导优于未萌动种胚。 The effect of seed sterilization conditions, various basal media, and growth regulators on explants of mature Taxus chinensis var. mairei embryos were studied. Results showed optimal germination occurred with seeds soaked in a 25.0 g·L-1 sodium hypochlorite solution for 10 min in a vacuum. The contamination rate was reduced to 6% and the germination rate reached 86%. A woody plant medium (WPM) was selected as the best basal medium with 0.1 mg·L-1 kinetin (KT) and 1.0 mg·L-1 zeatin (ZT). This medium effectively induced more clusters of adventitious bud formation; whereas 6-benzylaminopurine (6-BA) and isopenlenyladenine (2iP) induced fewer adventitious buds while N-phenyl-N′-1, 2, 3-thidiazol-5-ylurea (TDZ) produced no adventitious bud differentiation. With WPM, germinated embryos effectively induced adventitious buds.
