橙皮甙抑制高脂血症大鼠肝脏C反应蛋白分泌

摘要 目的观察橙皮甙对高脂血症大鼠血清中C反应蛋白(CRP)含量、肝组织CRP mRNA及蛋白表达的影响。方法将42只SD大鼠随机分为7组(每组6只):对照组、模型组、低、中、高剂量橙皮甙组(低、中、高剂量组)、辛伐他汀组(他汀组)、橙皮甙+辛伐他汀联合组(联合组)。对照组予普通饮食,其他6组均喂饲高脂饮食,按分组灌胃1次/d,持续1 2周。实验6、12周时检测血清CRP、TC、TG的含量。1 2周处死大鼠,采用RT-PCR法、Westernblot法检测大鼠肝脏CRP mRNA及蛋白的表达情况。结果与对照组比较,模型组大鼠6、1 2周CRP、TC和TG明显升高(P<0.01),与模型组比较,低剂量组CRP、TC和TG无显著差异(P>0.05);中、高剂量组、他汀组、联合组CRP、TC和TG显著降低(P<0.01),联合组较他汀组和高剂量组差异更显著。与对照组比较,模型组大鼠肝脏CRP mRNA及蛋白表达量明显升高(P<0.01);与模型组比较,中、高剂量组、他汀组、联合组肝脏CRPmRNA和蛋白显著降低(P<0.05);联合组较高剂量组、他汀组更显著(P<0.01)。结论橙皮甙降低高脂血症大鼠血清CRP的机制与其抑制肝脏CRP mRNA及蛋白的表达有关,且抑制效应呈剂量依赖性。 Objective To observe the effects of hesperidin(HPD) on the levels of C-reactive protein(CRP) in hyperlipemia(HL) rats’ blood sera and the expression of CRP mRNA and protein in HL rats’ liver.Methods SD rats were fed with high fat diet and given different concentrations of HPD or simvastatin(SV) through gavage for 90 days.The contents of CRP,TC and TG in blood serum were detected after 6 weeks and 12 weeks.CRP mRNA and protein in rats’ liver were detected by RT-PCR and Westernblot.Results The levels of CRP,TC and TG in model group significantly increased compared with control group(P〈0.01).The levels of CRP,TC and TG in moderate and high dose of HPD group,SV group and HPD+SV(HSV) group decreased compared with those in model group(P〈0.01).The decrease was more significant in HSV group than in SV group and high dose of HPD group(P〈0.01).The expression of CRP mRNA and protein in liver of rats in model group significantly increased compared with that in control group(P〈0.01) and had no significant difference compared with low dose of HPD group(P〉0.05).The expression of CRP mRNA and protein in liver of rats decreased significantly after treatment with moderate and high dose of HPD,SV and HSV compared with model group,and the decrease was more signifiant in HSV group than in high dose of HPD and SV groups(P〈0.01).Conclusion HPD can decrease the level of CRP,TC and TG and the mechanism of which might be related to the inhibition of the expression of CRP mRNA and protein in liver.
