枸杞多糖对大鼠坐骨神经离断后神经生长因子表达的影响 被引量:3

摘要 目的研究枸杞多糖对大鼠坐骨神经离断后神经生长因子(NGF)表达的影响。方法雌性SD大鼠72只,随机分成空白对照组、模型组与枸杞多糖(LBP)组,每组24只,后两组切除右侧坐骨神经2cm,LBP组腹腔注射枸杞多糖10mg/(kg.d),模型组给予相同剂量生理盐水,分别于给药后第7、14、21和28d取坐骨神经离断的近端神经及相连背根神经节和相对应的脊髓(每组6只),应用免疫组织化学方法分析神经生长因子表达的变化。利用Image-proplus6.0图像分析系统进行半定量分析。结果模型组和LBP组的坐骨神经及相连背根神经节和相应的脊髓NGF免疫组化平均光密度高于空白对照组,LBP组坐骨神经、脊神经节及脊髓中的NGF免疫组化平均光密度在术后第7、14、21、28d均低于模型组组(P<0.05)。结论枸杞多糖可抑制大鼠坐骨神经离断后近端及相应的节段神经节和脊髓组织中的神经生长因子量的表达。 Objective To investigate the effect of Lycium barbarum polysaccharides( LBP) on expression of nerve growth factor ( NGF) after sciatic nerve mutilation in rats. Methods 72 female SD rats were randomly divided into three groups: Control group, Model group and Lycium Barbarum Polysaccharides( LBP) group,Model group and LBP group were wipe off 2cm right sciatic nerve,LBP group were intraperitoneal injected LBP 10mg/( kg.d) ,Model group were given physiological saline,Control group were treated without any treatment. Six rats were choosed randomly and the near-heart never of sciatic nerve and conjoint dorsal root ganglia and spinal cord were obtained on the7th,14th,21st and 28th day; Immunohistochemistry and Image-pro plus 6. 0 were used to evaluate the expression of NGF. Results The expression of NGF positive area and Average optical density in Control group are obviously lower than the other group. The NGF Average optical density in sciatic nerve,dorsal root ganglia and spinal Cord in LBP group are obviously lower than that in Model group on the 7th,14th,21st,28th day( P0. 05) . Conclusion LBP can inhibition the expression of NGF the related nerve ganglion and spinal cord after sciatic nerve divided in rats. expression of NGF owned in sciatic nerve and its related nerve ganglion and spinal cord after sciatic nerve dividing in rats.
