摘要
建立了以2,4-二硝基氟苯(DNFB)作衍生化试剂,0.015mol/L磷酸氢二钠和磷酸二氢钾缓冲液和50%乙腈水溶液为流动相,采用反相HPLC法测定烟叶中20种游离氨基酸含量的方法,并采用该法测定了7个烟叶样品。结果表明:①20种氨基酸的平均回收率在94.2%~118.4%之间;相对偏差小于3%;②4个烤烟样品中均未检出甲硫氨酸和异亮氨酸,均是脯氨酸含量最高,且2个上部烟叶样品中的游离氨基酸总量都大于2个中部烟叶样品;③7个烟叶样品中,唯有巴西白肋烟BCRA总氨基酸含量最高,其天门冬氨酸的含量显著地高于其他烟叶样品;④马里兰烟苯丙氨酸的含量在7个烟叶样品中是最高的,且其天门冬酰胺和脯氨酸含量显著地高于其它6个烟叶样品;希腊香料烟样品中的脯氨酸略低于马里兰烟。
A method for determining 20 free amino acids in tobacco leaf was established by using 2,4 - dinitrofluorobenzene (DNFB) as a derivative reagent, 0.015 mol/L disodium hydrogen phosphate and potassium dihydrogen phosphate as buffer solution and 50% aqueous acetonitrile as mobile phase, and reversed phase HPLC. Seven tobacco leaf samples were detected. The results showed that: 1 ) The average recoveries of the 20 amino acids were from 94.2% to 118.4% with the RSDs of less than 3%. 2) Methionine and isoleucine were not detected in four flue-cured tobacco samples, the content of proline was the highest, and the contents of total free amino acids in two upper leaf samples were higher than those in two cutter samples. 3) The content of total free amino acids in the sample of Brazil burley tobacco, BCRA, was the highest, and its aspartic acid content was significantly higher than that in other samples. 4) The phenylalanine content in the sample of Maryland tobacco was the highest, and asparagine content was obviously higher than that in other samples, and proline content was slightly higher than that in the sample of Greece oriental and significantly higher than that in the other five samples.
