摘要
目的本研究通过对胃癌及癌前病变组织中Dnmt3A和Dnmt3B表达水平的检测,为探讨其在胃癌发病中的作用机理提供依据。方法通过手术及内镜活检取68例病例标本,依据病理检查结果,分为三组:胃癌组26例;胃癌前病变组(萎缩性胃炎伴肠化及不典型增生)20例;正常对照组21例。应用免疫组织化学SP法检测组织中Dnmt3A和Dnmt3B的表达水平。结果胃癌组Dnmt3A表达的阳性率是96.2%,胃癌前病变组是65.0%,正常对照组是76.2%。在胃癌组织中Dnmt3B表达的阳性率96.2%,胃癌前病变组是55.0%,正常对照组是81.0%。结论 1.胃癌组织中Dnmt3A、Dnmt3B高表达。Dnmt3A、Dnmt3B可能通过促进DNA甲基化,抑制相关基因的表达,在胃癌发生发展的过程中,Dnmt3A、Dnmt3B可能参与胃癌的分化过程。2.检测胃黏膜组织Dnmt3A、Dnmt3B可能是一种有用的肿瘤标志物。3.Dnmt3A、Dnmt3B的表达不参与胃癌的进展。
Objective:Objective By investigating the expression of Dnmt3A and Dnmt3B in gastric cancer and its premalignant lesions to offer evidence in gastric cancer generating..Methods: Immunohistochemistry was used to detect the expression of Dnmt3A and Dnmt3B in 26 patients with gastric cancer,20 patients with premalignent lesions and 25 cases of controls with superficial gastritis.Results: The positive rate of expression of Dnmt3A in patients with gastric cancer was 96.2%,while the premalignent lesions was 65.0% and persons with superficial gastritis was 84.0%.The positive rates of expression of Dnmt3B in patients with gastric cancer was 96.5% while the premalignent lesions was 55.0%,and persons with superficial gastritis was 80.0%.Conclusion: 1.It was showed that the high expression of Dnmt3A and Dnmt3B in gastric cancer tissues.Dnmt3A and Dnmt3B may be involved in carcinogenesis of gastric cancer by inducing genomic DNA hypermethylaton that was associated with inactive anti-oncogene.Dnmt3A and Dnmt3B may be involved in differentiation of gastric cancer.2.Dnmt3A and Dnmt3B in patients with gastric cancer tissue may be a useful tumor marker.3.Dnmt3A and Dnmt3B may be not associated with progression of gastric cancer.
