强脉冲光照射对成纤维细胞分泌TGF-β_1的影响及信号机制研究 被引量:4

Impacts of intense pulsed light on transforming growth factor-beta1 secretion in cultured human fibroblasts and the signaling mechanism

摘要 目的研究强脉冲光(intense pulsed light,IPL)照射对皮肤成纤维细胞分泌TGF-β1的影响及信号机制。方法分离培养皮肤原代成纤维细胞,分为两组:一组为IPL治疗组,IPL照射能量密度分别为0、10、18、27、36和36J/cm2×2(36J/cm2的IPL重复治疗2次),采用Elisa法检测细胞培养上清液中TGF-β1的含量,Western印迹法检测JNK和P38的磷酸化;另一组为IPL+抑制剂组,包括IPL对照,IPL+SP600125(JNK抑制剂),IPL+SB203580(P38抑制剂),在加入抑制剂2h后,36J/cm2的IPL照射,48h后检测培养上清液中TGF-β1的含量。结果 IPL照射后,IPL治疗组的TGF-β1的浓度在10、18、27和36J/cm2时分别与阴性对照组相比较均减少,而在36J/cm2×2时与阴性对照相比较增高;IPL治疗组中JNK和P38可被激活,随剂量增加,先增强后减弱,都在10J/cm2表达最强。IPL+抑制剂组上清液中TGF-β1的含量,IPL+SP600125与对照相比较减少(P<0.05);IPL+SB203580与对照相比有明显增多(P<0.05)。结论强脉冲光在较低能量密度(≤36J/cm2)时抑制皮肤成纤维细胞TGF-β1的分泌,较高能量密度(36J/cm2×2)时能促进TGF-β1的分泌;蛋白激酶JNK、P38在IPL影响TGF-β1的分泌过程中可能起着重要作用,其中JNK促进其分泌,而P38抑制其分泌。 【Objective】To study the impacts of Intense Pulsed Light(intense pulsed light,IPL) On transforming growth factor beta-1 secretion in cultured human fibroblasts and the signaling mechanism in the process.【Methods】 The callan foreskin fibroblasts were cultured and then divided into two groups.The first group,the IPL treatment group,was irradiated with IPL with fluences of 0 J/cm2(negative control),10,18,27,36,36 J/cm2×2(irradiated with IPL with fluences of 36 J/cm2 twice),then transforming growth factor beta-1(TGF-β1)were evaluated using enzyme-linked immunosorbent assay and P-JNK(phosphorylated,JNK) and pP38(phosphorylation,P38) were evaluated using western blot.The second group,IPL+inhibitor group,including the IPL(control),IPL+SP600125(JNK inhibitor),IPL+SB203580(P38 inhibitor),was irradiated with IPL with fluences of 36 J/cm2 two hours after adding the inhibitor SP600125 and SB203580.TGF-β1 in cultural supernatants was evaluated using enzyme-linked immunosorbent assay.【Results】 In the IPL treatment group:the concentrations of the TGF-β1 in cultural supernatants of 10,18,27,36 J/cm2 were reduced compared with the negative control,respectively,while in 36 J/cm2×2,increased;both JNK and P38 were activated after the irradiation of IPL,the erpression of them first increased and then decreased with the fluences increasing,while the strongest expression was in 10 J/cm2.In the IPL pluses inhibitor group:the concentration of the TGF-β1 in cultural supernatants of IPL +SP600125 reduced compared with the control,while in IPL + SB203580 increased(P 0.05).【Conclusion】 Intense pulsed light could suppress the secretion of TGF-β1 at the lower fluences(≤36 J/cm2),while could promote it at higher fluence(36 J/cm2×2).Protein kinases JNK,P38 may play opposite roles in the process of IPL affecting the secretion of TGF-β1,in which JNK promots its secretion and P38 inhibits its secretion
