摘要
目的研究视网膜光化学损伤中前列腺素(prostaglandin,PG)含量变化以及地塞米松(dexamethasone,DXM)对这种变化的影响。方法36只SD大鼠分为对照组和DXM组,后者于光照前3天开始,每日腹膜腔注射DXM(1mg/kg),连续5天。两组大鼠经(1900±106.9)lx,波长为510~560nm的光连续照射24小时,于光照前及光照结束后的6小时和1,3,7,14天分别摘除眼球,用放射免疫分析法进行视网膜前列腺素E2(prostaglandinE2,PGE2)与6-酮-前列腺素F1α(6-keto-PGF1α)含量检测。结果对照组光照后6小时,1,3天PGE2的含量为(37.50±2.75,48.06±4.04,81.90±4.89)pg/mg,6-keto-PGF1α的含量为(4.68±0.69,7.50±0.57,10.40±0.71)pg/mg,明显高于DXM组相应时间点的PGE2(20.60±4.28,37.36±3.34,54.85±4.57)pg/mg及6-keto-PGF1α(2.50±0.59,4.68±0.81,6.87±1.10)pg/mg(P<0.01)。结论DX?
Purpose To evaluate the prostaglandins(PG) levels
and to identify the effect of dexamethasone(DXM) on PG in response to photochemical insult in
rat retina. Methods The experiments were performed on 36 SD rats which were separated into
two groups,control and treated groups,and the latter received daily intraperitoneal injections of
DXM (1mg/kg)for 5consecutive days,starting 3 days before light exposure.The animals were
continually exposed to green fluorescent light(510560 nm)with an illuminance level of (1900106.
9)lx for 24 hrs.The retinal concentration of PGE2 and 6ketoPGF1
weretestedat6hrs,1,3,7and14daysafterlightexposure. Results ThePGE2and6ketoPGF1levels of
the control groups (37.502.75,48.064.04,81.904.89) pg/mg and
(4.680.69,7.500.57,10.400.71)pg/mghad significantly higher values than those of the treated
rats(20.604.28,37.363.34,54.854.57)pg/mg and (2.500.59,4.680.81,6.871.10)pg/mg( P <0.01)after
6hrs,1and 3 days light exposure respectively. Conclusion By inhibition of PG
synthesis,the DXM may play an ameliorative effect on retinal photochemical injury of rats.
