Determination of Psoralen and Isopsoralen in Shengfa Tincture by High Performance Liquid Chromatography

摘要 目的建立生发酊质量标准。方法用薄层色谱法对处方中的补骨脂、闹羊花定性鉴别;高效液相色谱法定量分析处方中补骨脂素、异补骨脂素的含量。结果薄层色谱法鉴别了补骨脂、闹羊花;色谱柱:CAPCELLPAKC18(250mm×4.6mm,5μm),流动相:甲醇-水(50:50);检测波长:246nm;流速:1.0ml/min。补骨脂素在6.00~60.00μg范围内呈良好的线性关系该,线性回归方程为:Y=73364.32X-3.59,r=0.9999;异补骨脂素在6.56~65.64μg范围内呈良好的线性关系,线性回归方程为:Y=73454.85X+4.01,r=0.9999,平均回收率补骨脂素为101.17%,RSD为0.96%;异补骨脂素为99.49%,RSD为1.75%。结论该方法准确、灵敏度高,重复性好。 Objective To set up a quality standard for Shengfa Tincture.Methods Fructus Psoraleae and Naoyanghua in the prescription were qualitatively identified by thin layer chromatography.The octadecylsilane bonded column was used as the stationary phase,and methanol-water (50:50) as the mobile phase.The column was CAPCELL PAK C18(250 mm×4.6 mm,5 μm),with a detection wavelength of 246 nm and a flow rate of 1.0 ml/min.Results The calibration curve of psoralen was linear over the range of 6.56-65.64 μg,and the equation of linear regression was Y=73 364.32X-3.59(r=0.9999).The calibration curve of isopsoralen was linear over the range of 6.56-65.64 μg,and the equation of linear regression was Y=73 454.85X+4.01(r=0.9999),The average recovery of psoralen and isopsoralen was 101.17%(RSD=0.96 %) and 99.49% (RSD=1.75%),respectively.Conclusion The method is reliable and accurate.
