摘要
目的观察电针对围绝经期抑郁症模型大鼠的效应。方法采用双侧卵巢切除(Ovariectomy,OVX)、孤养与慢性不可预知的温和应激(Chronic Unpredictable Mild Stress,CUMS)两步法建立围绝经期抑郁症的复合模型。造模后运用两组组穴(内关、后三里穴及关元、三阴交穴)进行电针,同时设立正常组、围绝经期抑郁症模型组和抑郁症模型组进行对照,测定大鼠Open-field行为学评分、糖水消耗量、体重变化、血清雌二醇(E2)、黄体生成素(LH)、促肾上腺皮质激素(ACTH)和皮质酮(CORT)含量变化。结果与正常组大鼠比较,围绝经期抑郁症模型组大鼠Open-field行为学评分、糖水消耗量明显下降,体重明显增加,血清E2、ACTH含量明显降低(P<0.05);与复合模型组比较,电针组大鼠Open-field行为学评分明显增加(P<0.05),糖水消耗量显著增多(P<0.01),同时血清E2、ACTH、CORT含量增加,体重下降,但差异无统计学意义(P>0.05)。结论电针可增强围绝经期抑郁症模型大鼠活动度,增加探究活动;改善慢性应激导致的快感缺乏。同时对该复合模型大鼠血清E2、ACTH、CORT含量的降低和体重增加均有良性调整趋势。
Objective To investigate the effect of electroacupuncture in a rat model of perimenopausal depression.Methods A composite model of perimenopausal depression was made by two steps:bilateral ovariectomy,and individual feeding and chronic unpredictable mild stress.After model building,electroacupuncture was performed at two groups of points(Neiguan(PC 6) and Housanli(ST 36),and Guanyuan(CV 4) and Sanyinjiao(SP 6)).Normal,perimenopausal depression model and depression model groups were established as controls.The Open-field behavior score was counted,and sugar water consumption,body weight and serum estradiol(E2),luteinizing hormone(LH),adrenocorticotrophic hormone(ACTH) and corticosterone(CORT) contents were measured in the rats.Results The Open-field behavior score,sugar water consumption and serum E2 and ACTH contents decreased and body weight increased significantly in the perimenopausal depression model group of rats compared with the normal group(P 0.05).The Open-field behavior score and sugar water consumption increased significantly(P0.05 and P0.01 respectively),and serum E2,ACTH and CORT contents increased and body weight decreased,which had no statistically significant difference(both P 0.05),in the electroacupuncture group of rats compared with the composite model group.Conclusion Electroacupuncture can strengthen movements,increase searching activities and improve anhedonia due to chronic stress in a rat model of perimenopausal depression.It also has a favourable tendency to regulate decreased serum E2,ACTH and CORT contents and increased body weight in the rat composite model.
